Here's what I used to do (advice from a star bassist back in the '40s):
I would first coat the blister with iodine, then I would burn the tip of a sewing needle and puncture the blister and let it drain, then coat the area with iodine again. I would coat again with iodine before going to the gig.In a couple of days it would begin to itch and playing becomes such a great pleasure. That's the beginning of the callous formation.
